- AbstractResidual and endangered populations of silver fir in air-polluted areas of the Ore Mountains lend themselves to ecological-genetic investigations. With respect to low degree of genetic diversity in Ore Mountains silver fir populations the investigations focus on: 1. parentage analyses in selected populations with natural regeneration. Parentage analyses are assumed to retrospectively allow conclusions on precedent mating events, including disturbances of mating system. 2. genetic comparison of silver fir from selected European populations of a broad geographic range. Results are expected to enlarge the basis for discussion of loss of genetic diversity in Ore Mountains silver fir populations. Genetic investigations to 1. and 2. shall be performed on the basis of DNA-polymorphisms. Common methodical approaches to detect DNA-polymorphisms are presented and discussed with regard to their suitability for parentage analyses and/or assessment of genetic diversity among populations. First results of RFLP- and PCR-based analyses of silver fir DNA are presented.
