- AbstractTwo populations of European silver-fir (Abies alba Mill.) from the natural range of the species in Sudety Mts were investigated isoenzymatically at 16 loci. These two populations are located at the eastern edge of a huge, man made, disjunction in silver fir range. The disjunction is generated by lethal for silver-fir, emissions of Czech, German, and Polish industry. To describe the genetic variation and genetic structure of populations we analyzed 16 enzymatic loci: Est-C, Fle, Gdh, Got-A, Got-B, Idh-A, Idh-B, Lap-A, 6-Pgdh-A, 6- Pgdh-B, Pgi-A, Pgi-B, Pgm-A, Pgm-B, Sod-A, Sod-B. Almost half (47%) of the investigated loci were polymorphic, at criterion 99%. Average observed and expected heterozygosity amounted to 0.134 and 0.126 respectively with average numbers of alleles per locus equal to N.=1.75. The calculated fixation index F= -0.122 for the more vigorous population Ladek Zdroj, revealed a small excess of heterozygosity. The second population, Miedzygorze with F= +0.006 is almost within the Hardy-Weinberg equilibrium. Multilocus outcrossing rate estimated for two polymorphic loci (Pgd-A and Idh-B) in Ladek Zdroj and four loci (6-Pgd- A, 6-Pgd-B, Idh-A and Idh-B) in the Miedzygorze embryo populations showed very low value of indices with t.=0.421 (0.0) for Ladek Zdroj, and 0.339 (+/-0.065) for Miedzygorze. Differences in t. value between the investigated populations are probably connected to the spatial structure of the stands. In both studied populations silver-fir is an admixed species.
