- AbstractArmillaria isolates from Zimbabwe were characterised by morphological features and isozyme analysis. Cultural characteristics on 2% malt extract agar (2% MEA), 3% MEA +peptone and on potato dextrose agar (PDA) showed three distinct groups designated I, II and III. The isolates also separated into the same three groups based upon rhizomorph patterns obtained in soil and also upon the ability of the isolates to fruit and type of fruit bodies obtained on oranges, millet ans wood inocula. The characteristics of fruit bodies obtained in the wild and in vitro for groups I and III, differed from both Armillaria heimii and Armillaria mellea. The morphological grouping was supported by isozyme analysis of pectic enzymes, Beta-1,3 glucanases, esterases and protein patterns obtained by electrophoresis and isoelectric focusing in polyacrylamide gels. European Armillaria species differ both morphologically and in isozyme patterns from Zimbabwean Armillaria. It can be concluded that Zimbabwean Armillaria are composed of three morphological and molecular groups and that pectic enzymes and glucanases might be useful in delineating taxa in Armillaria.
