Abstract
This article deals with the evalution of effect of growth regulators on in vitro formation, multiplication, and rooting of shoots of Spanish chestnut (Castanea sativa Mill..). Our results support the concept that the formation, multiplication, and rooting of shoots is dependent not only on the concentration and the type of growth regulators but also on the place of taking and on the duration of the cultivation. The multiplication intensity is also influenced by the explant position on the shoot and the date when it was exicised. When the explants were taken from the upper part of the twigs and the terminal bud was not removed, the number of shoot increased and the apical dominancy was very marked. The rooting ability was affected by the shoot length and by the duration of the cultivation. More adult material produced increased volume of polyphenoles, necrotized and died. At the concentration of 1.00 and 2.00 mg 1 hoch -1 IBA in cultivated agar medium the shoots of Spanish chestnut rooted.
