- AbstractBuds pre-treated with 50 uM benzyladenine (BA) and shoots of four willow clones (Salix spp.) cultured in vitro on hormone-free medium and on medium with 1 uM BA were analysed for cytokinins using an enzyme linked immunosorbent assay with polyclonal antibodies against isopentenyladenosine (iPA) and zeating riboside (ZR). The clone which was most difficult to root yielded the highest ZR titers. A clone with poor growth and a tendency to become necrotic had very high iPA titers, possibly indicating uptake of BA to a toxic level, since BA cross-reacts with antibodies against iPA. Other genotypic differences relating to axillary shoot production, shoot elongation growth and vitrification could not be correlated to cytokinin contents.
