Abstract
One option for gene conservation in forest trees would be the storage of pollen. At present this can be reactivated only in combination with female flowers. It would bei much more interesting if plants could be regenerated directly from pollen. This study has the aim to regenerate plants from early pollen stages and to store embryos in liquid nitrogen. Embryos of oak could be regenerated from pollen by anther culture. For Quercus petraea we found a practicable way to store globular and heartshaped embryos in liquid nitrogen using the method of SAKAI (1985) which we modified for ohr purpose (Abb.1). The embryos have been regenerated on a modified MS-medium (MURASHIGE and SKOOG; 1962) or a WPM (LLOYD and McCOWN, 1980) with 0 to 10 uM/1 BAP and 2,5 to 10 uM/1 2,4-D. The temperature was 28-30 degrees C and the light intensity 2000 lux (Abb.2).
