Abstract
Discriminating characters which reflect underlying genetic differences between the European intersterility groups (ISG) of Heterobasidion annosum occur within the internal transcribed spacer (ITS) and the intergenic spacer (IGS) of the ribosomal RNA gene repeat. RFLPs of the ITS rDNA repeat allow ISG-P to be unequivocally identified from both ISG-S and ISG-F. Discrimination between ISG-S and ISG-F isolates was developed using single strand DNA conformational polymorphism (SSCP) of the ITS locus or an amplification- refractory probing system. Restriction of PCR amplified fragments of the NTS of the ribosomal repeat could distinguish between ISG-P isolates. Polymorphic repeat elements were detected in the Heterobasidion annosum genome by human minisatellites or the M13 repeat. DNA bands that are common within each ISG were detected from a Wurope wide collection of Heterobasidion annosum isolates. Minisatellite banding allows identification of ISG-S and ISG-F, independent of the ribosomal SSCPs and restriction polymorphisms.
