Abstract
Various explants of white spruce (Picea glauca (Moench) Voss) seedlings and mature embryos were evaluated for survival following freezing to liquid nitrogen temperature (-196C). Cotyledons from nongerminated embryos which had been cryoprotected using a combination of 0.4 M sorbital, 5% DMSO, and 2% sucrose were frozen, employing a droplet-freezing technique, at an optimal cooling rate of 0.5C.min-1 to -40C prior to immersion in liquid nitrogen. Under these conditions, up to 35% of the cryopreserved cotyledons regenerated shoots upon return to in vitro culture. Some of the regenerated shoots were successfully rooted.
