Abstract
In this study, we investigated the usage possibilities of detached branches and needles in pathogenicity and virulence studies of Ophiostoma ulmi and Lophodermium pinastri. For this aim, isolation, diagnosis and purification of isolates, determination of pathogenicity, inoculation to detached shoots and needles were carried out for each pathogens under laboratory conditions. In order to determine the relationship between pathogenicity and relative water content, relative water content of elm shoots was determined. Inoculation to elm shoots and pine needles and measurement of relative water content of elm shoots were repeated monthly from April to October. Consequently, convenience of methods for each pathogens was demonstrated. It was determined that, there is a significant difference between months for pathogenicity of Ophiostoma ulmi. May and June are the most suitable months for inoculation to elm branches. Inoculation of L. pinastri to the needles may be done from April to October in laboratories.
