- AbstractFor the determination of cytokinins in spruce needles, Iyophilized needles were extracted with ethanol, and the cytokinins were pruifiedby DEAE-cellulose chromatography followed by Sep-Pak C18 cartridge extraction. After HPLC separation, isopentenyladenine, isopentenyladenosine, t-zeatin and t-zeatin riboside were quantified in the fractions by an indirect enzyme-linkedimmunosorbent assay. The recovery was almost 70% or more, dependent on the typ e of cytokinin. Estimations of cytokinins in spruce needles gave higher concentrations for t-zeatin riboside and isopentenyladenosine than for the corresponding free bases. A comparison of the cytokinin contents of differently damaged trees of two stands showed higher levels of cytokinin ribosides in 2 year-old needles of strongly damaged trees (e.g. t-zeatin riboside: 264 and 384 pmol/g dry weight; isopentenyladenosine: 437 and 702 pmol/g dry weight) compared with healthy or slightly damaged trees (t-zeatin riboside: 72 and 94 pmol/g dry weight; isopentenyladenosine: 281 and 314 pmol/g dry weight).
